Analysis of rice microbial communities under different storage conditions using culture-dependent and -independent techniques

Rice is a staple food for over half of the world’s population, and fungal spoilage in stored rice may occur when moisture content temperature are conducive. Aspergillus sp. Penicillium most harmful toxigenic species that produce mycotoxins. Molds pose potential threat to public health cause huge economic loss. Therefore, it great importance find out how molds multiply rice. This study focused on isolation identification fungi presented their evolution with different contents varying periods time under temperatures. Mold community was detected every month using culture-dependent -independent method polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). Significant differences were by traditional culture storage conditions. For potato dextrose agar (PDA) media, high suitable dominant strains including aurantiogriseum oxalicum. In particular, P. oxalicum competitively inhibited other fungi. Rose Bengal medium, no difference observed conditions, only typical such as candidus Alternaria detected. PCR-DGGE identified some uncultured Trichoderma Cladosporium sp., flora diversity restrictus Eurotium athecium. These results indicated conditions greatly shape growth. provides foundation during China help developing biological control methods prevent mold contamination
